serosal n-butyric acid (O, O,1, 1, or 1O mmollL, adjusted to neutral pH) on the epithelial cell proliferation rate of pig colonic mucosa in organ culture taking crypt cell production rate (CCPR) as the measure of proliferative activity With O or O.1 mmolfL n-butyric acid on the serosal side, luminal n-butyric acid increased CCPR at 1,O mmol/L, and decreased CCPR at 1O mmollL when compared to the Iuminal O mmolfL control, With 1.0 or 1O mmolfL serosal n-butyric acid, luminal n-butyric acid depressed CCPR dose-dependently The above results indicated that n-butyric acid stimulated colonic epithelial cell proliferation at low concentration and inhibit it at high concentration with interaction effect to enhance the inhibitory action. The stimulatory effect of a low dose of serosal n-butyric acid may be

K+ channel KVLQT1 located in the basolateral membrane of distal colonic epithelium is not essential for activating Cl- secretion.

The cellular mechanism for Cl(-) and K(+) secretion in the colonic epithelium requires K(+) channels in the basolateral and apical membranes. Colonic mucosa from guinea pig and rat were fixed, sectioned, and then probed with antibodies to the K(+) channel proteins K(V)LQT1 (Kcnq1) and minK-related peptide 2 (MiRP2, Kcne3). Immunofluorescence labeling for Kcnq1 was most prominent in the lateral ...

Mechanisms underlying distension-evoked peristalsis in guinea pig distal colon: is there a role for enterochromaffin cells?

The mechanisms underlying distension-evoked peristalsis in the colon are incompletely understood. It is well known that, following colonic distension, 5-hydroxytryptamine (5-HT) is released from enterochromaffin (EC) cells in the intestinal mucosa. It is also known that exogenous 5-HT can stimulate peristalsis. These observations have led some investigators to propose that endogenous 5-HT relea...

Substance P-evoked Cl(-) secretion in guinea pig distal colonic epithelia: interaction with PGE(2).

Interaction between substance P (SP) and PGE(2) on Cl(-) secretion in the guinea pig distal colonic epithelia was investigated. A short-circuit current (I(sc)) was measured as an index of ion transport. Mucosa preparations deprived of muscle and submucosa of distal colon were mounted in the Ussing flux chamber and treated with TTX and piroxicam to remove the influences of neuronal activity and ...

Melatonin inhibits tachykinin NK₂ receptor-triggered 5-HT release from guinea pig isolated colonic mucosa.

BACKGROUND AND PURPOSE Melatonin is involved in the regulation of colonic motility, and sensation, but little is known about the influence of melatonin on 5-hydroxytryptamine (5-HT) release from colonic mucosa. A tachykinin NK₂ receptor-selective agonist, [β-Ala⁸]-neurokinin A(4-10) [βAla-NKA-(4-10)] can induce 5-HT release from guinea pig colonic mucosa via NK₂ receptors on the mucosal layer. ...

Abnormal pattern of mucus-associated M1 antigens in histologically normal mucosa adjacent to colonic adenocarcinomas.

Apparently normal mucosae adjacent to colon adenocarcinomas were studied by cutting strips of mucosa from the entire length of 120 surgical specimens (94 located on the distal colon and 26 on the proximal colon). These mucosae were coiled into "Swiss rolls." Their mucus alterations were mapped by immunoperoxidase using antibodies against M1 antigens, oncofetal markers associated with precancero...
